The study was aimed to screen out the excellent tobacco germplasm resources which could be used in the field of scientific research, production and breeding. Conventional chemical composition and major alkaloids were tested, compared and clustered for 141 flue-cured tobacco, 18 burly tobacco and 6 cigar germplasm resources from China and abroad. Total sugar and reducing sugar contents were high in flue-cured tobacco germplasms, while total nitrogen, nicotine and potassium oxide were high in burley tobacco germplasms. Total alkaloid and nicotine content were the highest in flue-cured tobacco germplasm Zilaihuang 2243, and the content of cigar germplasm Dutch (ohio) was the lowest. The nicotine and nicotine conversion rates were the highest in the tobacco germplasm Pingbanliu, and those for tobacco germplasm Erxingzi and Burley11B was the lowest. The 165 accessions were divided into 4 groups for clustering analysis. Flue-cured tobacco germplasms were mainly concentrated in Group 1 and 4, characterized by high total and reducing sugar content and low nicotinic conversion rate. Group 2 was low in total and reducing sugars, and high in total nitrogen, total alkaloids and potassium oxide. Group 3 had low nicotinoids and total alkaloids, high nicotinic reduction, and high nicotinic conversion rate. The chemical composition and alkaloids of different germplasm resources have significant difference. The selected germplasms with different gradients could provide useful material for further study of nicotine.

为筛选能为科研、生产和育种利用的优异烟草种质资源。对来源于国内外的141份烤烟、18份白肋烟和6份雪茄烟的烟叶常规化学成分和主要生物碱进行检测、比较和聚类分析。结果表明:总糖和还原糖含量以烤烟最高,总氮、烟碱和氧化钾以白肋烟种质最高。总植物碱和烟碱含量以烤烟种质自来黄2243最高,雪茄烟种质Dutch(Ohio)最低;降烟碱和烟碱转化率以烤烟种质平板柳叶最高,以烤烟种质二性子和白肋烟种质Burley11B最低。聚类分析将165份种质分成4类,烤烟种质主要集中在第1和第4类,其主要特征是总糖和还原糖含量较高,烟碱转化率较低;第2类的总糖和还原糖含量低,总氮、总植物碱和氧化钾含量高;第3类的烟碱和总植物碱含量低,降烟碱含量高,烟碱转化率高。不同类型种质资源的化学成分和生物碱差异明显,筛选出的不同梯度烟碱的种质可为烟碱的进一步研究提供材料支撑。

为筛选能为科研、生产和育种利用的优异烟草种质资源。对来源于国内外的141份烤烟、18份白肋烟和6份雪茄烟的烟叶常规化学成分和主要生物碱进行检测、比较和聚类分析。结果表明:总糖和还原糖含量以烤烟最高,总氮、烟碱和氧化钾以白肋烟种质最高。总植物碱和烟碱含量以烤烟种质自来黄2243最高,雪茄烟种质Dutch(Ohio)最低;降烟碱和烟碱转化率以烤烟种质平板柳叶最高,以烤烟种质二性子和白肋烟种质Burley11B最低。聚类分析将165份种质分成4类,烤烟种质主要集中在第1和第4类,其主要特征是总糖和还原糖含量较高,烟碱转化率较低;第2类的总糖和还原糖含量低,总氮、总植物碱和氧化钾含量高;第3类的烟碱和总植物碱含量低,降烟碱含量高,烟碱转化率高。不同类型种质资源的化学成分和生物碱差异明显,筛选出的不同梯度烟碱的种质可为烟碱的进一步研究提供材料支撑。

The study was aimed to screen out the excellent tobacco germplasm resources which could be used in the field of scientific research, production and breeding. Conventional chemical composition and major alkaloids were tested, compared and clustered for 141 flue-cured tobacco, 18 burly tobacco and 6 cigar germplasm resources from China and abroad. Total sugar and reducing sugar contents were high in flue-cured tobacco germplasms, while total nitrogen, nicotine and potassium oxide were high in burley tobacco germplasms. Total alkaloid and nicotine content were the highest in flue-cured tobacco germplasm Zilaihuang 2243, and the content of cigar germplasm Dutch (ohio) was the lowest. The nicotine and nicotine conversion rates were the highest in the tobacco germplasm Pingbanliu, and those for tobacco germplasm Erxingzi and Burley11B was the lowest. The 165 accessions were divided into 4 groups for clustering analysis. Flue-cured tobacco germplasms were mainly concentrated in Group 1 and 4, characterized by high total and reducing sugar content and low nicotinic conversion rate. Group 2 was low in total and reducing sugars, and high in total nitrogen, total alkaloids and potassium oxide. Group 3 had low nicotinoids and total alkaloids, high nicotinic reduction, and high nicotinic conversion rate. The chemical composition and alkaloids of different germplasm resources have significant difference. The selected germplasms with different gradients could provide useful material for further study of nicotine.

烟草中生物碱达50余种,其中最主要是烟碱、降烟碱、假木贼碱和新烟草碱等[1],其含量烟碱>新烟草碱>降烟碱>假木贼碱[2-3]。烟碱是烟草中重要的生物碱,正常情况下,烟碱含量占总生物碱含量的93%以上[4],降烟碱含量一般不超过总生物碱含量的3%[5]。降烟碱极易发生亚硝化反应生成亚硝酸和烟草特有亚硝基降烟碱(NNN)[6],随着降烟碱含量的升高,NNN含量呈直线增加[7],烟碱转化导致降烟碱含量增高而影响烟叶的品质和安全性[8],而随着烟碱转化程度的提高,白肋烟风格下降,香气量变少,香气质变劣,杂气增加,主要香气物质含量降低[9]。研究发现,烟碱转化率与烟碱含量呈极显著负相关,而与降烟碱含量呈极显著正相关;新烟碱含量和假木贼碱含量与烟碱转化率无显著相关关系[10];不同类型的烟草、不同的研究材料其总生物碱、烟碱和降烟碱含量有差异。其中,总生物碱和烟碱含量晒烟>白肋烟>烤烟>马里兰烟>香料烟,降烟碱的含量和烟碱转化率以白肋烟最高,烤烟最低,香料烟、马里兰烟和地方晒烟均存在烟碱转化问题,降烟碱也有烤烟较高的情况[11-17]。我国国家烟草种质库目前已编目烟草资源5267份[18],云南也收集保存并编目烟草资源3076份,编写出版了《烟草种质资源图鉴》,编入各类型烟草资源1477份[19],本项目选择来自中国主要烟叶产区省份和美国、古巴等国家的165份3个类型烟草种质资源进行分析研究,以找出不同烟草类型、不同品种和不同来源地资源的生物碱差异,并筛选出优异资源为烟草育种、科研和生产提供材料支撑。
从云南省烟草农业科学研究院种质库中保存的3076份烟草种质资源中,按5%左右的比例抽取不同产地的资源,主要来自中国河南、山东、山西、安徽、贵州、黑龙江、辽宁、福建等省份及美国和古巴等国家的烟草种质资源165份,其中,烤烟141份、白肋烟18份、雪茄烟6份(表1)。试验位于云南省玉溪市研和镇的云南省烟草农业科学研究院试验基地,采用随机区组设计,重复3次,每个小区20株,行距1.20 cm,株距0.50 cm,每公顷施纯氮97.5 kg,N∶P2O5∶K2O=1.0∶0.5∶3.0,18份白肋烟种质资源施肥量加倍;种植土壤为砂壤土,pH 6.41,含有机质11.20 g/kg、全氮0.56 g/kg、全磷1.03 g/kg、全钾6.45 g/kg,有效氮84 mg/kg、有效磷91 mg/kg,有效钾162 mg/kg。按优质烟种植技术规范进行管理,成熟烟叶分小区采收,编秆后统一进行烘烤和晾晒调制,取中部烟叶各种质重复混合烟叶样品,送云南三标农林科技有限公司检测常规化学成分(总糖、还原糖、总氮、烟碱、水溶性氯、氧化钾、淀粉)和主要生物碱(烟碱、降烟碱、新烟碱、假木贼碱)等。
参照YC/T 159—2002测定水溶性糖含量,参照YC/T 166—2003测定总蛋白质含量,参照YCT 160—2002测定总植物碱含量,参照YC/T 173—2003测定钾含量,参照YC162—2002测定氯含量,参照GB/T 5009.9—2003测定淀粉含量,采用气相色谱/质谱分析烟草中的主要生物碱。按以下公式计算烟碱转化率:烟碱转化率=[降烟碱/(烟碱+降烟碱)]×100%。
采用Excel软件进行数据统计分析和作图,采用DPS软件进行聚类分析。
表2可知,总糖和还原糖含量最高的为烤烟种质湄潭黑团壳,雪茄烟种质Havana 503b和白肋烟种质Blue Star100最低;总植物碱含量以烤烟种质自来黄2243最高,雪茄烟种质Dutch(Ohio)最低;氧化钾含量以雪茄烟种质Havana 503b最高,烤烟种质湄潭黑团壳最低。生物碱中的烟碱、降烟碱、新烟碱、假木贼碱等含量最高、最低值均为烤烟种质,烟碱转化率以烤烟种质平板柳叶最高,白肋烟种质Burley11B最低。变异系数除降烟碱和烟碱转化率较高(218.23%和262.74%),为强变异性,其余成分的CV为17.13%~80.40%,均为中变异性。
3种类型的烟草种质资源的化学成分中,总糖、还原糖和总植物碱含量以烤烟最高,雪茄烟和白肋烟较低;总氮和氧化钾含量以白肋烟种质为高,总氮含量烤烟和雪茄烟较低,总植物碱含量以雪茄烟Dutch(Ohio)最低,氧化钾含量以烤烟最低,淀粉含量以烤烟最高,雪茄烟和白肋烟较低。变异系数以雪茄烟的总植物碱含量最高(106.31%),为强变异性,其余成分为中变异性。化学成分的最大值、最小值、平均值、变异系数等不同类型烟草种质间有差异,白肋烟种质的总氮含量变异系数较小(9.61%),为弱变异性,雪茄烟种质总植物碱含量的变异系数较大(106.31%),为强变异性,其余各项为中变异性;各类烟草各化学成分的极值及其对应种质中,湄潭黑团壳、小黄金0003、Burley2、GR10、Greider等种质有2项以上的最大值或最小值(表3)。
141份烤烟种质烟叶的化学成分差异较大。其中,总糖含量较高(>24.00%)的7份,在18.00%~24.00%适宜范围的33份;总氮含量除黑苗2308、黑苗白筋2338、竖把柳叶2110、自来黄2243>3.50%外,其余种质均在2.01%~3.50%适宜范围;总植物碱含量>5.00%的2份,4.01%~5.00%的4份,3.01%~4.00%的23份,<1.50%的38份,其余种质在1.50%~3.5%之间;氧化钾含量为4.01%~5.00%的5份,3.01%~4.00%的18份,2.01%~3.00%的83份(表4)。
18份白肋烟种质资源总糖和还原糖含量较低,分别为0.53%~2.10%和0.30%~1.79%;总氮含量较高(3.47%~5.21%);氧化钾含量较高(3.59%~ 5.32%);总植物碱含量差异较大(0.68%~4.64%)。其中,Burley37的总植物碱含量为4.64%,Burley49、Burley11B、Burley21(V246-Rhodesia)、GR18、GR10等种质的烟碱含量均在3.01%~4.00%之间,Burley1、GR14、Ex.12、GR10A等种质总植物碱含量较低(0.68%~1.55%),其余种质烟碱含量在2.01%~3.00%之间;氧化钾含量Burley64、GR10A、GR14等种质的氧化钾含量>5.00%,Burley10A、Burley2、GR18、Blue Star100、GR36、Burley1、Burley11B、GR17B、Burley37、GR10、Ex.12、GR17等种质在4.01%~5.00%之间,其余种质在3.59%~4.00%之间(表5)。
6份雪茄烟种质资源总糖和还原糖含量较低,总氮含量适中(2.08%~3.64%),总植物碱和氧化钾含量差异较大,分别为0.41%~4.37%和1.58%~6.08%。其中,总植物碱含量以Greider最高,Dutch(Ohio)最低;氧化钾含量以Havana 503b最高,Greider最低(表6)。
不同类型烟草烟碱含量范围含量为烤烟为0.99~44.12 mg/g、白肋烟3.74~ 37.40 mg/g、雪茄烟1.01~35.42 mg/g;烟碱平均含量以白肋烟最高,烤烟次之,雪茄烟最低;降烟碱均值以雪茄烟最高,烤烟次之,白肋烟最低;各类烟碱转化率较高的种质为烤烟平板柳叶、雪茄烟Dutch(Ohio),白肋烟Burley1;烟碱转化率均值以雪茄烟最高,烤烟次之,白肋烟最低;各类型烟叶的烟碱转化率、降烟碱和雪茄烟的烟碱等变异系数均>100%,为强变异性,其余成分为中变异性。各类烟草各化学成分的极值及其对应种质中,Burley37、Ex.12、Greider、Havana 503b、Dutch(Ohio)、自来黄2243、二性子等种质有2项以上的最大值或最小值(表7)。
白肋烟种质烟碱含量在30.01~40.00 mg/g之间的有3份(Burley11B、Burley37、Burley49),20.01~ 30.00 mg/g的有8份,10.00~20.00 mg/g的6份,<10.00 mg/g的1份(Burley1);烟碱转化率以Burley1最高,为52.98%,Burley37为6.33%,其余种质为0.28%~1.56%(表8)。
雪茄烟种质资源的烟碱含量以Greider为最高,Dutch(Ohio)最低,其余种质在0.99~20.00 mg/g之间;烟碱占比Dutch(Ohio)最低为18.67%,Havana 501为79.42%,其余种质在97.95%~98.54%之间;烟碱转化率以Dutch(Ohio)最高,为80.69%,Havana 501为19.58%,其余种质≤1.33%,甚至低至0.64%(表8)。
141份烤烟种质资源中,烟碱含量以自来黄2243最高,小黄金0003最低;松边黄苗榆2202、晋太207、黑苗白筋2338、小尖梢、小黄金0029、竖把柳叶2116、黑柳子等种质烟碱含量在30.01~40.00 mg/g之间,有100份种质的烟碱含量<20.00 mg/g。烟碱转化率在0.37%~89.64%之间,小白筋2515最低,平板柳叶最高;平板柳叶、小黄金0003、泼拉机、葵花烟2437、黔南9号、黑苗2319、尖烟洋苗、武鸣4号等种质烟碱转化率>50.00%,黑苗2344、黑苗烟2343、大竖把2141等种质在20.01%~50.00%之间,烤烟、湄潭铁杆烟、竖把2135、圆叶稠码、小竖把2146、小黄金0091、晋太75、黑苗2341、晋太207、麻江柳叶烟等在5.01%~20.00%之间(表8)。
史志宏等[10]按烟碱转化率将烟叶分为4组:非转化株(<5%)、低转化株(5%~20%)、中高转化株(20%~50%)和高转化株(>50%)。按此标准将165份资源进行分类,并列出各转化类型的种质资源名单(烤烟21份,白肋烟和雪茄烟各2份)(表9)。
为提高可比性和资料的可靠性,分别对不同来源的烤烟和雪茄烟类型资源进行比较分析,白肋烟为同一来源(美国),不作比较。
图1可以看出,不同来源烤烟资源的化学成分和烟碱转化率差异较大。各成分平均值中总糖和还原糖含量以来源于中国贵州的资源最高,中国黑龙江的资源最低(图1A);总植物碱以来自中国安徽的资源最高,氧化钾含量以中国贵州的资源最高(图1B)。烟碱、降烟碱、烟碱转化率等均值以中国安徽的资源最高,辽宁的最低(图1B图1C);不同来源种质资源的总糖、还原糖、烟碱转化率、烟碱、降烟碱等多个成分最大值差异较大,最小值以总糖、还原糖、总植物碱等差异较大,其余成分差异较小(图1D图1E)。变异系数以降烟碱差异最大(图1F)。
雪茄烟种质资源来自美国和古巴,统计结果表明,各化学成分平均值中以总氮、氧化钾含量以古巴资源较高(图2A);总生物碱和烟碱转化率以美国资源较高(图2B图2C)。各化学成分的最大值和最小值差异不明显(图2D);而烟碱、降烟碱含量和烟碱转化率的最大值美国资源明显高于古巴(图2E);变异系数在1.37%~285.07%之间,其中,达到强变异性(CV≥100.00%)的有:美国的总糖和还原糖;中国山西和山东以及古巴和美国的降烟碱;中国安徽、贵州、河南和山东以及古巴和美国的烟碱转化率等。CV在10.0%以内的有中国福建、贵州、黑龙江、辽宁和山东的总氮;黑龙江的新烟碱和假木贼碱和辽宁的氧化钾等;其余来源及化学成分的变异系数均在10.00%~99.99%之间,为中变异性(图2F)。
将165份烟草种质资源的总糖、还原糖、总氮、总植物碱、水溶性氯、氧化钾、淀粉、烟碱、降烟碱、新烟碱、假木贼碱含量和烟碱转化率等12项指标,采用标准化转化、欧氏距离、离差平方和法进行系统聚类,在截距为0.9左右处将165份资源分成4类(图3),每一类均可以再分为2个亚类,各类种质数量及编号见表10,其差异性比较和各成分的变异系数见图4,各类及其主要特征特性为:
第1类:包含78份种质,可分成2个亚类。其中,1-1亚类63份均为烤烟种质;1-2亚类中烤烟、白肋烟和雪茄烟种质分别为8份、3份和4份。其主要特征是总糖和还原糖含量较高,烟碱转化率较低。
第2类:包含28份种质,可分成2个亚类。其中,2-1亚类中有12份烤烟和1份雪茄烟种质;2-2亚类中白肋烟有14份和烤烟1份(竖叶子0982)。其主要特征是总糖和还原糖含量低,总氮、总植物碱和氧化钾含量高。
第3类:包含10份种质,3-1亚类有烤烟种质4份;3-2类中分别有烤烟4份、白肋烟和雪茄烟种质各1份。其主要特征是烟碱含量低,总植物碱含量低,降烟碱含量高,烟碱转化率高。
第4类:包含烤烟种质49份,均为烤烟种质。其主要特征是总糖、还原糖和淀粉含量高,总氮含量低。
按以上同样的聚类方法对141份烤烟资源进行聚类分析,在0.9左右处将141份资源分成4类,每一类均可分成2个亚类(图5),各类所包含的种质资源数量及编号见表11,各类主要特征特性为:
第1类氧化钾含量高;第2类总糖、还原糖、淀粉、总植物碱、烟碱含量低,降烟碱含量高,烟碱转化率高;总植物碱、烟碱含量高;第3类总植物碱、烟碱含量最低,降烟碱含量和烟碱转化率最高;第4类总糖、还原糖、淀粉、总植物碱、烟碱含量较高:第5类的总糖、还原糖、淀粉含量高,总氮含量低。第1、4类的降烟碱含量和烟碱转化率的变异系数较高(≥100%),为高变异性,各类的总氮、第5类的总糖含量等为弱变异性,其余的为中变异性(图6)。
烟碱是烟叶中主要的生物碱,其含量在生物碱中占95%以上,烟碱含量随烟草类型、品种、产地、施肥等的不同而有差异。本研究结果表明,不同类型的烟草及同一类型不同品种间的烟碱含量差异较大,其范围为烤烟的烟碱含量为0.99~ 44.12 mg/g、白肋烟为3.74~37.40 mg/g、雪茄烟为1.01~35.42 mg/g。而有研究显示香料烟为2.33~ 5.19 mg/g、白肋烟为17.36~25.50 mg/g和烤烟为23.66~24.400 mg/g[20],本研究多样性更丰富。孙鑫等[21]从219份烤烟资源中筛选出烟碱含量高的品种福泉朝天立、8022、开阳团鱼叶、毕金一号和长葛柳叶等资源,许美玲等[22]从52份烤烟资源中筛选出烟碱含量>5.00%的小黄金0009和小黄金5209。本研究筛选出烟碱含量高(>40.00 mg/g)的种质自来黄2243(烤烟)和Burley37(白肋烟)等;烟碱含量在30.00~40.00 mg/g之间的烤烟种质8份(松边黄苗榆2202、晋太207、黑苗白筋2338、小尖梢、小黄金0029、黑柳子、竖把柳叶2116、胎里富1060)等、白肋烟种质2份(Burley49、Burley11B)和雪茄烟种质Greider。这些种质可作为高烟碱资源进行进一步研究和利用。
按烟碱转化率的高低可将烟草分为高转型、中高等转化型、低转化型和非转化型种质[11]。本项的165份种质中,高转型种质有烤烟8份(平板柳叶、小黄金0003、泼拉机、葵花烟2437、黔南9号、黑苗2319、尖烟洋苗、武鸣4号),占5.67%;白肋烟Burley1(占5.55%)和雪茄烟Dutch(Ohio)(16.67%)等;中高等转化型种质烤烟3份(黑苗2344、黑苗烟2343、大竖把2141)。低转化型种质雪茄烟Havana 501和烤烟种质偏筋黄1036、晋太3号、晋太6号、黄苗2218、小黄金0029、小尖梢等。田阳阳等[23]研究的TS01株系的烟碱转化率在93%以上,证明烤烟的烟碱转化率也较高;雪茄烟品种均存在烟碱向降烟碱的转化问题,烟碱转化率范围为5.03%~53.12%[24],国内外不同产区雪茄烟叶和茄芯、茄衣等不同用途雪茄烟叶生物碱含量样本间差异性均较大,特别是烟碱、降烟碱含量和烟碱转化率变异性极大,表明雪茄烟叶烟碱向降烟碱转化问题突出[25],本研究结果与以上文献基本相符。另外,SHI等[5]和BUSH等[26]的研究认为,美国白肋烟品种在群体中一般含有15%~20%的转化株,约1/2转化株的烟碱转化率>20.00%。本研究认为,由于烟草类型不同、品种不同,烟碱含量及烟碱转化率差异较大。如平板柳叶、小黄金0003等烟碱转化率在85.00%以上,与硃砂烟(浅红、中等红色、深红3种类型硃砂烟叶的烟碱转化率分别为86.95%、90.86%、93.85%,远高于云烟87的5.12%)烟碱转化率相似[27]。烟碱转化率较高的资源可作为硃砂烟叶等特殊用途进行研究和利用。
3种类型的烟草种质资源总糖和还原糖含量以烤烟最高,雪茄烟和白肋烟较低;总氮、总植物碱和氧化钾含量以白肋烟种质为高,总氮含量以烤烟和雪茄烟较低,总植物碱含量以雪茄烟最低,氧化钾含量以烤烟最低,淀粉含量以烤烟最高,雪茄烟和白肋烟较低。有研究表明,不同基因型间烟叶含钾量有较大差异,而选择吸收和积累钾能力强的品种是最有效的措施[12],黄莺等[28]提出在常规施钾量水平下烤烟富钾、高钾等基因型的划分标准,用该标准对本研究的141份烤烟种质氧化钾含量进行划分的结果表明:富钾型(>2.50%)种质36份,占25.53%,高钾型(1.80%~2.50%)种质87份,占61.70%,低钾型(<1.80%)种质18份,占12.77%。其中,麻江立烟、黑苗2308、竖叶子0982、竖把大柳叶2133、黔南1号等5份的氧化钾含量>4.00%;黄苗2218、小白筋2516、黔南3号、黔南7号、黑柳子、掩心烟2441、武鸣4号、黔南9号、黑苗2344、黑苗2322、牡单79-2、核桃纹2475、竖叶子0987、三保险2440、小黄金0019、黑苗毛烟、大柳叶2020、晋太125-11等18份的氧化钾含量在3.00%~4.00%之间。培育富钾烟草品种可提高我国烟叶含钾量,从而达到烟叶降焦的目的,并能提高烟株抗逆、抗病性,改善烟叶品质[29]
综上所述,不同烤烟种质资源间的化学成分和生物碱的含量差异较大。经过分析比较,筛选出了一批化学成分适宜,氧化钾含量高,降烟碱含量或烟碱转化率较低的种质,也区分出了高转化型、中高转化型和低转化型的烤烟种质。相关分析结果提示了各化学成分指标间的相关关系,聚类分析将特性不同的种质归为不同的类型。
