There are about 69 species of Mangifera in the world, and 8 species are mainly distributed in China. The long history of mango cultivation in China has resulted in many natural hybrids with local characteristics. In the process of continuous coordinated development with the environment, the appearance of mango, such as branches, leaves and fruits, has changed to some extent, but its pollen has formed unique morphological characteristics while maintaining genetic conservatism and stability. In order to further study the taxonomic significance of the pollen morphology of mango plants and clarify the phylogenetic relationship between mango natural hybrids, this study carried out palynological research and genetic relationship analysis on mango germplasm based on the microscopic characteristics of pollen, and observed the pollen morphology and pollen exine ornamentation of 32 mango materials from two species of mango using scanning electron microscopy. On this basis, cluster analysis was carried out on six quantitative characters of pollen. The results showed that the pollen of 32 the mango germplasms belonged to N3P4C5 type. The polar view was 3-split triangle, and the red view was nearly elliptical. There were three germination grooves, which almost reached both ends; There were differences in the shape and size of pollen, the width of germination ridge, pollen ornamentation, the density and size of pollen outer wall mesh among the materials. Stripes or reticular ornamentation and pits were all over the surface of pollen grains. Most mango pollen belongs to reticular ornamentation. Mesh shapes were diverse, mainly circular and long, with rough ridges. Mesh distribution density varied among different materials. According to the cluster analysis of quantitative characters such as pollen grain size, pollen morphology, germination ridge width, mesh size and mesh density, the 32 mango germplasms could be divided into three groups when the similarity distance was 15. Mango resources from Southeast Asia, such as Machesu M. indica, Siam Mango M. indica, Ren He Carabao M. indica, were clustered into one group in the pedigree map, showing certain regional characteristics; Yun Nan Tu Mango M. indica, Bruce Mango M. indica and Amemwins No.26 M. indica with narrow germination ridge were divided into one type; M. persiciformis, collected M. persiciformis and common domestic M. indica. cultivation species were grouped into one group. The original wild species, Bai Hua M. indica, was grouped with No.1 M. persiciformis, No.2 M. persiciformis, No.3 M. persiciformis, and No.4 M. persiciformis. The resource had the characteristics of primitive and wild types. This study would provide palynological basis for the classification, genetic relationship and evolution of mango pollen morphology.

芒果属全球约有69个种,我国主要分布有8个种。我国悠久的芒果栽培历史形成许多具有地方特色的天然杂交种。芒果在不断与环境协调发展的过程中,枝叶、果实等外观形态发生了一定的变化,其花粉在保持遗传保守性和稳定性下却形成了独特的形态特征。为了深入研究芒果属植物花粉形态的分类学意义,理清芒果天然杂交种之间的系统发育关系,本研究基于花粉微观特征,对芒果属种质开展孢粉学研究及遗传关系分析。利用扫描电镜系统观察芒果属2个种32份芒果材料的花粉外部形态和花粉外壁纹饰等形态特征,在此基础上,对花粉6个数量性状进行聚类分析。结果表明:32份芒果种质花粉均属于N3P4C5型,极面观呈3裂三角形,赤面观呈近椭圆形,萌发沟孔为3条,萌发沟孔开裂度几乎到达花粉粒的两极;各材料的花粉形状大小、萌发脊宽窄、花粉纹饰、花粉外壁网孔密度和大小均存在差异;条纹状或网状纹饰和纹孔布满花粉粒表面,大多数芒果花粉属于网状纹饰,网孔形状多样,主要是圆形和长条形,纹脊粗糙,网孔分布密度在不同材料间存在差异。根据花粉粒大小、花粉形态、萌发脊宽、网孔大小和网孔密度等数量性状对参试材料进行聚类分析,32份芒果种质在相似距离为15时可分成三大类群,来自东南亚的马切苏、暹罗芒、仁和吕宋等芒果资源在谱系图中聚为一类,显示出一定的地域特征;萌发脊较窄的云南土芒、布鲁斯和象牙芒26号聚为一类;扁桃、收集的扁桃-芒与国内常见芒果栽培种聚为一类。原始的野生种白花本地芒与扁桃1号、扁桃2号、扁桃3号、扁桃4号聚为一组,该类资源具有原始型、野生型的特点。本研究可为芒果花粉形态的分类、亲缘关系和演化研究提供孢粉学依据。

芒果属全球约有69个种,我国主要分布有8个种。我国悠久的芒果栽培历史形成许多具有地方特色的天然杂交种。芒果在不断与环境协调发展的过程中,枝叶、果实等外观形态发生了一定的变化,其花粉在保持遗传保守性和稳定性下却形成了独特的形态特征。为了深入研究芒果属植物花粉形态的分类学意义,理清芒果天然杂交种之间的系统发育关系,本研究基于花粉微观特征,对芒果属种质开展孢粉学研究及遗传关系分析。利用扫描电镜系统观察芒果属2个种32份芒果材料的花粉外部形态和花粉外壁纹饰等形态特征,在此基础上,对花粉6个数量性状进行聚类分析。结果表明:32份芒果种质花粉均属于N3P4C5型,极面观呈3裂三角形,赤面观呈近椭圆形,萌发沟孔为3条,萌发沟孔开裂度几乎到达花粉粒的两极;各材料的花粉形状大小、萌发脊宽窄、花粉纹饰、花粉外壁网孔密度和大小均存在差异;条纹状或网状纹饰和纹孔布满花粉粒表面,大多数芒果花粉属于网状纹饰,网孔形状多样,主要是圆形和长条形,纹脊粗糙,网孔分布密度在不同材料间存在差异。根据花粉粒大小、花粉形态、萌发脊宽、网孔大小和网孔密度等数量性状对参试材料进行聚类分析,32份芒果种质在相似距离为15时可分成三大类群,来自东南亚的马切苏、暹罗芒、仁和吕宋等芒果资源在谱系图中聚为一类,显示出一定的地域特征;萌发脊较窄的云南土芒、布鲁斯和象牙芒26号聚为一类;扁桃、收集的扁桃-芒与国内常见芒果栽培种聚为一类。原始的野生种白花本地芒与扁桃1号、扁桃2号、扁桃3号、扁桃4号聚为一组,该类资源具有原始型、野生型的特点。本研究可为芒果花粉形态的分类、亲缘关系和演化研究提供孢粉学依据。

芒果属  /  花粉形态  /  电镜扫描  /  聚类分析

There are about 69 species of Mangifera in the world, and 8 species are mainly distributed in China. The long history of mango cultivation in China has resulted in many natural hybrids with local characteristics. In the process of continuous coordinated development with the environment, the appearance of mango, such as branches, leaves and fruits, has changed to some extent, but its pollen has formed unique morphological characteristics while maintaining genetic conservatism and stability. In order to further study the taxonomic significance of the pollen morphology of mango plants and clarify the phylogenetic relationship between mango natural hybrids, this study carried out palynological research and genetic relationship analysis on mango germplasm based on the microscopic characteristics of pollen, and observed the pollen morphology and pollen exine ornamentation of 32 mango materials from two species of mango using scanning electron microscopy. On this basis, cluster analysis was carried out on six quantitative characters of pollen. The results showed that the pollen of 32 the mango germplasms belonged to N3P4C5 type. The polar view was 3-split triangle, and the red view was nearly elliptical. There were three germination grooves, which almost reached both ends; There were differences in the shape and size of pollen, the width of germination ridge, pollen ornamentation, the density and size of pollen outer wall mesh among the materials. Stripes or reticular ornamentation and pits were all over the surface of pollen grains. Most mango pollen belongs to reticular ornamentation. Mesh shapes were diverse, mainly circular and long, with rough ridges. Mesh distribution density varied among different materials. According to the cluster analysis of quantitative characters such as pollen grain size, pollen morphology, germination ridge width, mesh size and mesh density, the 32 mango germplasms could be divided into three groups when the similarity distance was 15. Mango resources from Southeast Asia, such as Machesu M. indica, Siam Mango M. indica, Ren He Carabao M. indica, were clustered into one group in the pedigree map, showing certain regional characteristics; Yun Nan Tu Mango M. indica, Bruce Mango M. indica and Amemwins No.26 M. indica with narrow germination ridge were divided into one type; M. persiciformis, collected M. persiciformis and common domestic M. indica. cultivation species were grouped into one group. The original wild species, Bai Hua M. indica, was grouped with No.1 M. persiciformis, No.2 M. persiciformis, No.3 M. persiciformis, and No.4 M. persiciformis. The resource had the characteristics of primitive and wild types. This study would provide palynological basis for the classification, genetic relationship and evolution of mango pollen morphology.

漆树科(Anacardiaceae)芒果属(Mangifera),约有69个种[1]。芒果在我国栽培历史悠久,传统栽培生产过程中存在许多天然杂交种,形成了野生、半野生和野生近源种等独具特色的地方资源。目前在芒果遗传多样性分析、指纹图谱构建等方面仍以SSR[2]、SRAP[3-4]及ISSR[4]等分子标记的手段分析为主,而分子标记本身受环境条件、标记类型等差异而导致分析结果不稳定。许文天等[1]和施宗明等[5]简要描述过花粉量及花粉形态特征,但未深入描述花粉超微形态特征,花粉形态指标且品种数量涉及范围也较小。芒果花粉超微结构在植物系统分类的研究报道较少。植物随着漫长的历史演变而不断与环境协调发展,其花粉在保持遗传保守性和稳定性下形成了独特的形态特征,国内外许多专家对柚[6]、枣[7]、菊芋[8]、悬钩子[9]、梨[10]等树种的花粉形态进行观察,对其品种的分类鉴定、起源演化等方面进行了研究。对芒果属植物进行聚类分析前,先把花粉形态学特征的观测数据采用数量化分析的手段进行结构化处理,该方式可成为人们对芒果属植物进行科学研究的一个新手段[11]。本研究对芒果属2个种32份种质资源花粉进行调查研究,可在孢粉学领域为芒果的系统分类、优特品种选育提供研究依据。
32份供试芒果花粉均于2021年采自广西亚热带作物研究所芒果种质资源圃(表1)。于盛花期随机采集树冠外围不同方向中上部雄蕊成熟的正常花朵,用灭菌镊子将成熟破裂的灰白色花药放入电镜固定液中,摇匀固定后,4 ℃低温保存备用。
花电镜观察方法参照方仁等[12]的方法,选择紫红色的花朵,将成熟的灰白色花粉进行脱水处理并干燥,用离子溅射仪对导电胶上紧贴的干燥花粉进行真空喷金镀膜、移入发射扫描电镜观察并拍照。每份芒果供试材料均选取具有代表性的视野观察记录8个花粉形态特征数据,每个特征3次重复。300倍观察花粉粒的群体形态,3000倍观察花粉的极面和赤道面,15 000倍观察花粉的外壁纹饰特征,并测量极轴长(P)、赤道轴长(E)、萌发脊宽度(W)及网孔直径(D)大小。花粉粒的形状用极轴长和赤道轴长的比值来表示(P/E),1.14<P/E≤2为长球形,0.88<P/E≤1.14为近球形,0.50<P/E≤0.88为扁球形,P/E≤0.5为超扁球形[13]
利用Excel软件计算平均值和标准差;选取极轴长、赤道轴长、萌发脊宽度、网孔直径、网孔密度值共5个定量特征用IBM SPSS 19.0软件的组间联接法进行系统聚类分析。
图1A~图1C可知,供试芒果花粉粒形状规则,大小匀称,均为3条花粉萌发孔沟,大部分花粉粒萌发沟孔开裂度几乎到达花粉粒的两极,萌发沟以等间距环状分布,萌发点凸起。对照NPC分类系统可知芒果花粉属于N3P4C5型。其赤道观大多数为椭圆形,萌发孔呈裂开状;其极面观为3裂三角形;条纹状或网状纹饰和纹孔布满花粉粒表面。芒果花粉的形状大小、萌发脊宽窄、网孔直径和密度存在不同程度的差异。32个芒果材料花粉除白花本地芒、百东扁桃-芒19-24花粉粒的沟孔在极面有轻微的汇合外,其他花粉粒沟孔在极面均不汇合。根据花粉性状P/E分级标准,云南土芒P/E值为1.56,属长球形花粉;扁桃1号、扁桃2号、东水一号、桂热芒23号、扁桃3号、土芒、扁桃4号、四季蜜芒和紫薇芒等9份材料花粉P/E值范围0.76~0.87,属扁球形花粉,其余的22份芒果材料属于近球形花粉。从赤道轴长看,云南土芒的最小,为15.05 μm,显著小于其他芒果资源。从萌发脊宽来看,象牙芒26号最小,为13.77 μm,与恒宁扁桃-芒19-3、百东扁桃-芒19-27、桂热芒82号、云南土芒、龙眼香芒、布鲁斯和秋实1号无差异,但显著小于其余24份芒果资源。从网孔密度来看,马切苏、暹罗芒网孔密度均为37 μm2,与百东扁桃-芒19-26、恒宁扁桃-芒19-3、百东扁桃-芒19-13、百东扁桃-芒19-12、百东扁桃-芒19-27、东水一号、象牙芒26号、龙眼香芒、帕拉英达、土芒、秋实1号、桂热芒289号、四季蜜芒、仁和吕宋和红花本地芒无差异,但显著大于其余15份芒果资源。
图1D可见,从花粉外壁纹饰来看,大多数芒果花粉属于网状纹饰,网孔形状多样,主要是圆形和长条形,纹脊粗糙,网孔分布密度在不同材料间存在差异。32个供试芒果种质材料花粉壁的表面纹饰都非常明显,但网纹特征存在一定的差异,且萌发脊宽窄不一。大致可将这32个芒果花粉壁的纹饰分为以下4类:(1)花粉粒极轴部呈网状纹饰,赤道面纹饰纵向成条纹状,与网纹结合。网孔呈圆形或者超长圆形,大小,形状差异大,网孔深。如白花本地芒(图1D-1)、扁桃1号(图1D-2)、扁桃2号(图1D-6)、云南土芒(图1D-19)、帕拉英达(图1D-21)、布鲁斯(图1D-22)、土芒(图1D-4)、秋实1号(图1D-25)、桂热芒289号(图1D-26)、扁桃3号(图1D-23)、扁桃4号(图1D-27)。(2)网纹呈脑纹状,纹路大而凸起,网孔较小,网孔密度稀疏,花粉粒表面粗糙,如金煌芒(图1D-5)、百东扁桃-芒19-24(图1D-8)、百东扁桃-芒19-16(图1D-9)、恒宁扁桃-芒19-3(图1D-10)、马切苏(图1D-16)、象牙芒26号(图1D-18)、紫薇芒(图1D-32)、仁和吕宋(图1D-30)、暹罗芒(图1D-28).(3)花粉粒极面和赤道面均为网纹状,网孔呈近圆形,大小相对均匀,如台农1号(图1D-3)、南逗迈4号(图1D-4)、龙眼香芒(图1D-20)、四季蜜芒(图1D-29)、桂热芒23号(图1D-17)、红花本地芒(图1D-31);条纹状纹饰呈交叉弯曲,纹脊粗大且连片,偶见纹孔。如百东扁桃-芒19-26(图1D-7)、百东扁桃-芒19-13(图1D-11)、百东扁桃-芒19-12(图1D-12)、百东扁桃-芒19-27(图1D-13)、桂热芒82号(图1D-14)、东水一号(图1D-15)。
根据表2中的极轴长/赤道轴长(P/E)、萌发脊宽、网孔密度等6个花粉形态特征数量性状,对32份芒果材料进行聚类分析。在相似距离为15时,32份芒果材料分为三大类群(图2)。
第一类包括马切苏、暹罗芒、仁和吕宋3份材料,这3份材料都是来自东南亚的品种,从聚类图来看,其亲缘关系较接近。脑纹,其网孔直径较小,网孔密度较大。第二类是云南土芒、布鲁斯和象牙芒26号,其萌发脊宽度较窄。其余的26份芒果资源归为第三类,其中扁桃1号、扁桃2号、扁桃3号和白花本地芒单独成组,该组是最原始的野生芒果,其花粉纹饰是网纹和条纹的结合;四季蜜芒、南逗迈4号和桂热芒23号聚为一组,其花粉粒纹饰为网纹纹饰,网孔近圆形,大小相对均匀。
携带大量遗传信息的花粉使植物在进化过程中保持较强的保守性和稳定性,为利用孢粉学进行遗传关系的分析提供了理论依据[14]。近年来属间及种间的分类研究工作已经在部分作物中展开,并已见利用孢粉学手段开展亲缘关系研究的报道。张梅等[15]对绣球属及近缘属花粉形态的研究表明绣球属的近缘种属花粉形态的变异范围被绣球属植物花粉形态的变异幅度覆盖了,且绣球属花粉特征性状在种内变异相对稳定,而种间差别明显。
本研究中参试的32份芒果种质资源分属2个种,其花粉在微观形态学上表现出一定的共性,其赤道观为圆形或椭圆形,呈裂开状;其极面观为3裂三角形;表面具脑纹状、条纹状或网状纹饰,属于N3P4C5型。但各芒果资源在花粉粒大小、萌发脊宽等指标存在着一定的差异,大小、深浅、疏密不等的穿孔分布在花粉粒表面。花粉粒大小与品种演化存在一定的关系,陈薇薇等[16]对当归属植物的研究显示花粉演化趋势经历了原始、中等进化和进化3种类型,花粉粒的极轴变长,形状由球形向超长球形演变。本研究通过电镜扫描观察发现,芒果属花粉粒主要为近球形或扁球形,只有云南土芒的花粉形状为长球形,这一现象与其研究不一致,可能是因为目前的芒果栽培种遗传关系复杂,是经过长年的自然杂交和人工杂交育种而得。
芒果属芒果种在我国已有上千年的栽培种植史,在多年的驯化引种过程中,可能存在亲缘关系背景杂合等现象,较近地区的大范围引种种植使得这些芒果种(M. indica L.)资源与本土的芒果属扁桃种(M. persiciformis C.)资源可能产生了自然杂交,存在适应性进化,形成了扁桃-芒。扁桃-芒的外观表型例如叶片呈现出扁桃种的表观性状,叶薄革质,狭披针形或线状披针形,长11~20 cm,宽2~2.8 cm[17]。但其在孢粉学形态上与芒果种更为接近。本研究中观察发现百东扁桃-芒19-13号与龙眼香芒聚在一起、桂热芒82号在遗传相似系数为3时与秋实一号聚在一起,同时与其聚为一类的还有百东扁桃-芒19-27、百东扁桃-芒19-26、百东扁桃-芒19-12,而桂热芒82号与秋实一号都是印度芒901的实生后代,这表明了扁桃种和芒果种之间可能不存在杂交障碍。
根据花粉粒大小、花粉形态、萌发脊宽、网孔大小和网孔密度等数量性状对参试材料进行聚类分析,在相似距离为15时,32份芒果属资源可分为三大类群。32个芒果属种质资源花粉形态特征具有一定的共性,同时来源地较为接近的芒果属种质资源在进化树上被聚为一类,例如马切苏、暹罗芒、仁和吕宋等东南亚来源地的芒果属资源划分特征较为一致,显示出一定的地域特征,在谱系图中聚为一类,这与采用SRAP分子标记手段进行分类的结果基本一致[3]。而白花本地芒、扁桃1号、扁桃2号、扁桃3号、扁桃4号的资源具有一定的相似性,该类资源具有原始型、野生型的特点。
芒果属资源的高度杂合背景使其花粉具有一定共性,为其分类和利用带来一定困难。因此,芒果种质资源花粉的电镜分析结果可以作为芒果鉴定分类的依据之一。但对从分类学的角度来看,单凭花粉形态对遗传背景高度复杂的芒果属资源进行数量化分类,还不能确定其分类地位,因此必须还要结合其各自的生物学特性对芒果属种质资源的鉴定和分类进行确认。总体而言,采用孢粉学手段,利用基因学、分子生物学和细胞学方法,结合芒果生物学性状进行综合分析揭示芒果属种质资源的亲缘演化关系,对高度进化的芒果属资源分类研究更有现实意义。
