In this paper, Miscanthus lutarioriparius (ML) cellulose was used as the raw material to produce bioethanol by a fedbatch synchronous saccharification and fermentation process. The effects of initial substrate concentration, final substrate concentration, and feeding times on ethanol yield were analyzed. The orthogonal experiments were used to optimize the process, and the optimal process parameters were obtained as follows: Initial substrate concentration was 17.5%, and the batch feeding was conducted twice until the final substrate concentration was 32.5%. Under these conditions, the highest ethanol content could reach to 70.15 g/L, which increased by 23.36% compared to the synchronous saccharification and fermentation process without fedbatch. The material balance analysis was conducted on the basis of the optimal process. The structure of ML cellulose before and after fermentation was characterized by scanning electron microscopy, fourier transform infrared spectroscopy, and Xray diffraction. The results showed that the enzymatic saccharification and fermentation process would change the internal structure of ML cellulose, break the hydrogen bond or covalent bond, and increase the crystallinity of the substrate.

文章以南荻纤维素为原料,采用分批补料同步糖化发酵工艺生产生物乙醇,分析了底物初始质量分数、底物最终质量分数、补料次数对乙醇产量的影响,并在此基础上采用正交试验进行工艺优化,获得最佳工艺参数:底物初始质量分数为17.5%,分批补料2次至底物最终质量分数为32.5%。在此条件下,乙醇含量最高可达70.15 g/L,较未补料同步糖化发酵过程获得的最大乙醇含量提高 23.36%。在分批补料同步糖化发酵最佳工艺的基础上,做了物料平衡分析,并采用扫描电镜、傅里叶变换红外光谱、X−衍射对发酵前、后的南荻纤维素结构进行表征,结果显示,酶解糖化及发酵过程会改变南荻纤维素的内部结构,使氢键或共价键断裂,底物结晶度提高。

In this paper, Miscanthus lutarioriparius (ML) cellulose was used as the raw material to produce bioethanol by a fedbatch synchronous saccharification and fermentation process. The effects of initial substrate concentration, final substrate concentration, and feeding times on ethanol yield were analyzed. The orthogonal experiments were used to optimize the process, and the optimal process parameters were obtained as follows: Initial substrate concentration was 17.5%, and the batch feeding was conducted twice until the final substrate concentration was 32.5%. Under these conditions, the highest ethanol content could reach to 70.15 g/L, which increased by 23.36% compared to the synchronous saccharification and fermentation process without fedbatch. The material balance analysis was conducted on the basis of the optimal process. The structure of ML cellulose before and after fermentation was characterized by scanning electron microscopy, fourier transform infrared spectroscopy, and Xray diffraction. The results showed that the enzymatic saccharification and fermentation process would change the internal structure of ML cellulose, break the hydrogen bond or covalent bond, and increase the crystallinity of the substrate.

随着我国经济的快速发展,能源消耗日益增加,能源安全危机凸显,能源安全是关系国家经济发展的全局性、战略性问题。生物燃料乙醇因生产原料资源丰富、可再生且绿色清洁等特点, 越来越受到各国能源机构的关注。南荻(Miscanthus lu-tarioriparius, ML)是我国特有的植物, 主要生长在洞庭湖流域,年产量约为 80 万 $\mathrm{t}$ ,其富含木质纤维素,尤其是纤维素占南荻干重的 40%,是生产生物质乙醇的优良原料。但是, 南荻原料结构致密,其纤维素酶解转化率低于 $5\%$ [ 1 ] ,需要一定的预处理以促进南荻的酶解糖化,从而提高后续发酵产乙醇效率。南荻原料经过甲酸处理后,可得到南荻纤维素,因其他组分的去除,纤维素相对含量增加,有助于可发酵糖的生成及后续发酵产率的提高 [ 2 ]
生产生物质燃料乙醇包含 5 大步骤, 分别是原料预处理、酶解糖化、乙醇发酵、蒸馏和精馏,影响生物乙醇产率的关键因素是纤维素糖化发酵工艺。纤维素乙醇糖化发酵工艺主要分为同步糖化发酵、分步糖化发酵、半同步糖化发酵等,其中同步糖化发酵因操作简便, 酶解糖化生成的可发酵糖能及时被酵母利用,减少了可发酵糖含量过高对纤维素酶活性及酵母活性的抑制等优点, 被认为是更适合工业化生产乙醇的糖化发酵工艺 [ 3 ] 。 发酵体系中乙醇质量分数的高低是生物质乙醇能否工业化生产的关键。有研究表明,当发酵体系的乙醇质量分数大于 4%时,能大大降低蒸馏成本 [ 4 ] 。 理论上, 底物质量分数与乙醇质量分数呈显著正相关;实际上,体系中底物质量分数过高会增大体系的粘度, 影响底物与酶、可发酵糖与酵母的接触,从而降低酶解和发酵效率 [ 5 ] 。有研究报道,在底物质量分数低于 15%的情况下, 木质纤维素发酵产乙醇的经济性较差, 原因在于酶解糖化所产生的可发酵糖无法满足高产发酵的要求 [ 6 ] ; 但有些吸胀性极强的原料,在底物质量分数为 15%时就会反向抑制体系的酶解糖化过程, 分批补料法能较好地解决以上问题。Zhao X B [ 7 ] 以甘蔗渣为原料, 采用分批补料提高体系的乙醇含量, 结果显示,乙醇含量最高可达 80 g/L。
本研究以经甲酸分离的南荻纤维素为原料, 采用单因素及正交试验研究分批补料同步糖化发酵产乙醇工艺并对其进行优化, 最终确定最佳工艺条件;采用扫描电镜(SEM)、傅里叶变换红外光谱(FT-IR)、X-衍射(XRD)等方法研究发酵前、后底物的微观形态和表观结构的变化, 以期为生物质乙醇的工业化生产提供技术支撑和理论基础。
南荻收集于岳阳市洞庭湖流域, 自然风干后, 粉碎过 40 目筛备用; 参照文献[ 8 ]提取及测定南荻木质纤维素(纤维素含量为 89.66%、半纤维素含量为 3.05%、木质素含量为 3.10%);酿酒酵母 (Saccharomyces cerevisiae,编号 CICC 33068)由中国工业微生物菌种保藏管理中心提供;甲酸、过氧化氢、氢氧化钠、乙醇、硫酸、葡萄糖、硫酸铵、磷酸二氢钾、无水氯化钙和无水硫酸镁(均为分析纯) 购于国药集团化学试剂有限公司; 酵母提取粉购于广州环凯微生物科技有限公司; D-葡萄糖和 D-木糖(标准品)购于 Sigma-Aldrich 公司;纤维素酶购于 Novozymes 公司。
Thermo Ultimate 3000 型高效液相色谱仪、 FW177 型粉碎机、TG16-Ⅱ 台式高速离心机、 BSA224S 型电子天平、HH-420 型恒温水浴锅、 DGH-9246A 型电热恒温鼓风干燥箱、YSQ-LS- 50S11 型立式压力蒸汽灭菌锅、ZHWY-2102C 型恒温培养振荡器、HZQ-F100 型恒温摇床、SW-CJ-2D型落地式洁净工作台、Eco-S15型纯水系统、Rigaku D/max 2500 型衍射仪、Hitachi S4800 型扫描电子显微镜、Thermo fisher Nicolet iS5 型红外光谱仪。
称取 $2\mathrm{\;g}$ 南荻纤维素至三角瓶中,加入 ${30}\mathrm{\;{mL}}$ 发酵培养基 [ 9 ] ,置于高压蒸汽灭菌锅中, ${115}^{\circ }\mathrm{C}$ 灭菌 30 min,取出,冷却至室温; 加入 20 FPU/g 纤维素酶(以底物质量计),并按 10%接种量加入酿酒酵母二代活化培养液,在 ${37}^{\circ }\mathrm{C},{150}\mathrm{r}/\mathrm{{min}}$ 条件下发酵 ${120}\mathrm{\;h}$ ;在 ${12},{24},{48},{72},{96},{120}\mathrm{\;h}$ 分别取上清液 ${0.2}\mathrm{\;{mL}}$ ,于 ${6000}\mathrm{r}/\mathrm{{min}}$ 下进行固液分离,测定上清液中的乙醇和葡萄糖含量。
①底物初始质量分数
在 1.3.1 的发酵条件下进行分批补料试验, 底物初始质量分数分别设为 $5\%,{7.5}\%,{10}\%,{15}\%$ , 底物最终质量分数均为 30%,补料次数为 3 次 (补料时间分别为 ${12},{24},{48}\mathrm{\;h}$ ),纤维素酶添加量 (以底物质量计)为 ${20}\mathrm{{FPU}}/\mathrm{g}$ ,在12,24,48,72,96, ${120},{144}\mathrm{\;h}$ 分别取上清液 ${200\mu }\mathrm{L}$ 进行固液分离, 测定葡萄糖和乙醇含量。
②底物最终质量分数
在 1.3.1 的发酵条件下进行分批补料试验, 底物最终质量分数分别设为 ${25}\%,{27.5}\%,{30}\%$ , 35%,底物初始质量分数均为 15%,补料次数为 3 次 (补料时间分别为 ${12},{24},{48}\mathrm{\;h}$ ),其他条件同上。
③ 补料次数
在 1.3.1 的发酵条件下进行分批补料试验, 补料次数分别设为1,2,3,4次,补料时间分别为 ${12},{24},{48},{72}\mathrm{\;h}$ ,底物最终质量分数均为 ${30}\%$ ,底物初始质量分数均为 15%, 其他条件同上。
在单因素试验结果的基础上, 以乙醇含量为指标, 采用 3 因素 3 水平正交试验对底物初始质量分数 (A)、底物最终质量分数 (B)、补料次数 (C) 进行优化, 以确定南荻纤维素分批补料同步糖化发酵最佳的工艺条件。正交试验因素及水平如 表 1 所示。
利用 Thermo Ultimate 3000 型高效液相色谱仪测定葡萄糖及乙醇含量。色谱仪配置及检测条件: 色谱柱为 Bio-RAD AminexR HPX-87H Ion Exclusion Column $\left({{300}\mathrm{\;{mm}}\times {7.8}\mathrm{\;{mm}}}\right)$ ; 流动相为 ${0.005}\mathrm{\;{mol}}/\mathrm{L}$${\mathrm{H}}_{2}{\mathrm{{SO}}}_{4}$ 溶液,流速为 ${0.6}\mathrm{\;{mL}}/\mathrm{{min}}$ ; 柱温为 ${55}^{\circ }\mathrm{C}$ ,柱后冷却温度为 ${30}^{\circ }\mathrm{C}$ ;检测器为示差折光检测器,检测器温度为 ${45}^{\circ }\mathrm{C}$ ; 进样体积为 10 $\mu {\mathrm{L}}_{\circ }$
将南荻纤维素原料及发酵残渣于 65 ℃下烘干粉碎, 然后进行扫描电子显微镜观察, 检测电压为 ${20}\mathrm{{kV}}$
将南荻纤维素原料及发酵残渣于 65 ℃下烘干粉碎,取适量样品与 $\mathrm{{KBr}}$ 研磨均匀后压片,在 ${4000}\sim {500}{\mathrm{\;{cm}}}^{-1}$ 波数内进行傅里叶转换红外光谱分析。
将南荻纤维素原料及发酵残渣于 ${65}^{\circ }\mathrm{C}$ 下烘干粉碎,然后进行 $\mathrm{X}$ -衍射光谱分析,采用连续扫描,扫描值为 $5 \sim {50}^{\circ }$ ,采样步宽为 ${0.02}^{\circ }$ 。样品结晶度 $C$ 的计算式为
$ C =\left({{I}_{002}- {I}_{\mathrm{{am}}}}\right)/{I}_{002}\times {100}\%$
式中: ${I}_{002}$${2\theta }= {18}^{\circ }$ 时的衍射强度; ${I}_{\mathrm{{am}}}$ 为主结晶峰 002 的最大衍射强度。
同步糖化发酵过程中底物质量分数对乙醇含量及转化率的影响如 图 1 所示。
图 1 (a) 可知:在底物质量分数低于 20% 的情况下, 随着底物质量分数的增加, 乙醇含量显著增加;当底物质量分数为 ${20}\%$ ,发酵时间为 ${72}\mathrm{\;h}$ 时,乙醇含量达到最大值56.87 g/L;当底物质量分数为 25%时,乙醇含量显著下降,特别是反应初期,当发酵时间为 ${12}\mathrm{\;h}$ 时,乙醇含量仅为 ${2.44}\mathrm{\;g}/\mathrm{L}$ , 随着发酵时间的延长,乙醇含量有所增加,但是相对于底物质量分数为 20% 的反应体系,乙醇含量下降了 54%~92.99%。
图 1 (b) 可知: 当底物质量分数为 ${10}\%\sim$ 20%时,体系的乙醇转化率均随着发酵时间的延长先增加后降低,且在发酵时间为 ${72}\mathrm{\;h}$ 时达到最大,此时乙醇转化率为 62.18%~70.80%,其中底物质量分数为 15%时的乙醇转化率最大,为 70.80%; 当底物质量分数进一步增加至 25%时,乙醇转化率显著下降至 20%左右。在一定的底物质量分数范围内, 通过提升底物质量分数能达到提高体系乙醇含量的目的, 但是, 当底物质量分数超过 20% 后,会出现负向效应。因此,本文拟通过分批补料来提升体系的乙醇含量,选取 15%为最大底物初始质量分数进行分批补料试验。
底物初始质量分数对葡萄糖和乙醇含量的影响如 图 2 所示。 图 2 中各体系均在 ${12},{24}\mathrm{\;h}$ 和 48 $\mathrm{h}$ 进行 3 次补料,补料后的底物最终质量分数均为 30%。
图 2 可知, 随着发酵时间的延长, 反应体系中的乙醇含量呈先增加后降低的趋势,葡萄糖含量呈持续增加的趋势。当发酵时间为 ${12}\mathrm{\;h}$ 时,底物初始质量分数为 $5\%,{7.5}\%,{10}\%,{15}\%$ 的反应体系的乙醇含量分别为 ${13.59},{17.55},{24.39},{31.12}\mathrm{\;g}/\mathrm{L}$ , 说明底物初始质量分数与乙醇含量存在显著正相关,原因是底物初始质量分数越高,酶解产生的可发酵葡萄糖含量越高,乙醇转化率也较高;此时仅在底物初始质量分数为 15%的体系中检出葡萄糖,含量为 ${0.39}\mathrm{\;g}/\mathrm{L}$ ,其他均为未检出。当发酵时间为 ${24}\mathrm{\;h}$ 时,各体系的乙醇含量均呈显著增加趋势,其增加率分别为 109.96%, 92.23%, 55.98%, 53.78%;此时,只在底物初始质量分数为 10% 和 15%的体系中检测出极少量葡萄糖。当发酵至 48 h,各体系的乙醇含量仍有显著提升,增加率分别为 69.65%, 53.36%, 31.31%, 18.82%; 此时,各体系中均积累了大量葡萄糖未被酵母完全利用。当发酵至 ${72}\mathrm{\;h}$ ,各体系的葡萄糖含量均显著增加, 而乙醇含量则有不同程度的下降, 原因可能是随着反应的进行,系统中的乙醇含量较高且残留大量葡萄糖,影响了酵母生长和发酵,使体系中的乙醇含量趋于平衡;另外,在发酵后期,酵母缺乏生长所需的碳源, 会利用部分乙醇作为碳源维持生长 [ 5 ] 。当反应时间进一步延长,各体系的葡萄糖含量有一定程度的增加, 但乙醇含量未有显著变化。
底物最终质量分数对葡萄糖和乙醇含量的影响如 图 3 所示。
图 3 可知, 随着发酵时间的延长, 反应体系的葡萄糖含量呈增加趋势, 乙醇含量先增加后稍有降低至动态平衡。4 个体系的底物初始质量分数均为 ${15}\%$ ,当发酵时间为 0~12 h时,酵母活性较高,体系中的乙醇含量显著上升;当发酵时间为 ${12}\mathrm{\;h}$ 时,4 个反应体系的葡萄糖含量均很低,乙醇含量相接近,为 ${32}\mathrm{\;g}/\mathrm{L}$ 左右,此时进行第 1 次补料,4 个体系分别添加底物 3.34%,4.17%,5.00% 和 ${6.67}\%$ 。当发酵时间为 ${24}\mathrm{\;h}$ 时,各体系的乙醇含量均显著增加,各体系的残留葡萄糖含量均较低, 说明酶解产生的葡萄糖均被酵母所利用, 此时进行第 2 次补料,补料量同第 1 次。当发酵时间为 ${48}\mathrm{\;h}$ 时,各体系的乙醇含量均达到最大值,分别为 ${57.33},{57.99},{61.37},{56.52}\mathrm{\;g}/\mathrm{L}$ ;此时,各体系均残留大量葡萄糖未被酵母利用,葡萄糖含量分别达到 ${3.15},{5.40},{11.22},{18.22}\mathrm{\;g}/\mathrm{L}$ ; 此时进行第 3 次补料, 使得 4 个体系的底物总质量分数分别达到 25%, 27.5%,30%和 35%。随着发酵时间的继续延长,各体系的乙醇含量趋于动态平衡, 葡萄糖含量均显著增加,且与底物总质量分数呈正相关。
理论上, 反应体系中底物质量分数越高, 其酶解糖化发酵产生乙醇的量越高。但是,底物最终质量分数为 35%的体系的最大乙醇含量为 4 个体系中最低,较底物最终质量分数为 30% 的体系减少了 7.91%,原因可能是底物质量分数超过一定范围后,底物吸胀导致反应体系中的粘稠度增大, 影响了酵母菌活力。在反应超过 ${48}\mathrm{\;h}$ 后,体系中的乙醇含量不增反减,原因可能是随着体系中乙醇和葡萄糖含量的不断增加,酵母的活性受到抑制;另外,随着发酵时间的延长,反应体系中的酵母会因为营养竞争等原因产生次生代谢产物, 酵母也逐步达到稳定期或衰退期,代谢产生的次生代谢产物, 如醛类、酸类等, 会在酵母分泌的酯类合成酶的作用下与乙醇发生反应, 使得乙醇含量下降[ 10 ]。
补料次数对葡萄糖和乙醇含量的影响如 图 4 所示。
(b)葡萄糖含量随发酵时间的变化
图 4 可知, 随着发酵时间的延长, 各反应体系的葡萄糖含量逐渐增加,乙醇含量呈先急剧增加再缓慢下降或稍有提升的趋势, 整体维持在一个平衡状态。当发酵时间为 ${12}\mathrm{\;h}$ 时,4个反应体系中的葡萄糖和乙醇含量接近,分别为 ${0.18}\mathrm{\;g}/\mathrm{L}$${30}\mathrm{\;g}/\mathrm{L}$ ,此时进行第 1 次补料,4 个反应体系中的底物总质量分数分别为 ${30}\%,{22.5}\%,{20}\%,{18.75}\%$ 。 当发酵时间为 ${24}\mathrm{\;h}$ 时,只有 1 次补料体系的葡萄糖含量显著增加,说明 1 次补料体系的葡萄糖出现了剩余;各体系的乙醇含量增长率分别为 51.27%, ${62.60}\%,{53.78}\%$${47.48}\%$ ,整体与补料量呈正相关, 仅 1 次补料体系例外, 原因可能是过高的底物质量分数会导致体系的粘稠度增加,不利于底物与酶充分接触, 使得酶解转化率降低, 影响可发酵葡萄糖的生成,进而影响乙醇产量。在第 24 小时进行第 2 次补料,当发酵到 ${48}\mathrm{\;h}$ 时,各体系中的乙醇含量均达到最大值,分别为 60.50,65.41, 57.88,59.59 g/L, ${24}\sim {48}\mathrm{\;h}$ 的乙醇含量增长率分别为 33.34%,31.11%,20.94%和 21.41%。在第 48 小时进行第 3 次补料, 此时 4 个反应体系中的葡萄糖含量均显著提升,但乙醇含量均有所下降。在第 72 小时进行第 4 次补料,此时各体系的底物最终质量分数均达到 30%,4 个反应体系中的葡萄糖含量均显著提升,但乙醇含量维持在较稳定的水平。
南荻纤维素分批补料同步糖化发酵条件优化的正交试验结果与分析如 表 2 所示, 方差分析如 表 3 所示。
表 2 可知, 3 个因素对发酵体系中乙醇含量高低的影响顺序为 $\mathrm{A}> \mathrm{C}> \mathrm{B}$ 。通过极差分析得到南荻纤维素分批补料同步糖化发酵的最佳工艺条件组合为 A3C2B3, 即底物初始质量分数为 17.5%,底物最终质量分数为 32.5%,补料次数为 2 次。
通过查询 $F -$ 分布表, ${F}_{0.05}\left({2,2}\right)= {19.00}$ , ${F}_{0.01}\left({2,2}\right)= {39.00}$ ,这表明底物初始质量分数和补料次数均对乙醇含量具有极显著影响, 而底物最终质量分数对乙醇含量没有显著影响。
根据 2.3.1 中正交试验结果筛选的最佳工艺参数 $\mathrm{A}3\mathrm{C}2\mathrm{B}3$ 进行 3 次验证试验,即底物初始质量分数为 ${17.5}\%$ ,在 ${12},{24}\mathrm{\;h}$ 后进行 2 次分批补料至底物最终质量分数为 32.5%, 按 1.3.1 中的试验方法进行分批补料同步糖化发酵试验, 所得最大的乙醇含量为 ${70.15}\mathrm{\;g}/\mathrm{L}$
以南荻为原料,通过甲酸分离,碱处理纯化, 获得南荻纤维素, 再采用 2.3.2 中优化所得的分批补料同步糖化发酵的方式制备乙醇, 相关步骤的物料平衡如 图 5 所示。
图 5 可以看出: ${1000}\mathrm{\;g}$ 南荻原料经甲酸及碱处理后,纤维素的回收率为 ${85.87}\%$ ; 后续分批补料同步糖化发酵,发酵时间为 ${48}\mathrm{\;h}$ 时取样测定,其中底物剩余量约为 33.54%, 其中纤维素剩余 ${94.36}\mathrm{\;g}$ ,说明此阶段酶解糖化的纤维素量达 76.46%,此部分纤维素酶解生成的葡萄糖一部分转化为乙醇,乙醇质量为 ${85.00}\mathrm{\;g}$ ,另有 ${110.19}\mathrm{\;g}$ 葡萄糖未被酵母利用。
南荻纤维素原料及发酵残渣的 SEM 图如 图 6 所示。
图 6 可知:酶解发酵前,南荻纤维素样品整体呈条状,排列不规则,结构松散、表面粗糙,有诸多球状、片状凸起,并伴有明显裂缝和孔洞;酶解发酵后, 南荻纤维素的表观形貌发生较大的改变, 纤维素整体仍呈条状,排列更加杂乱无章,但条状纤维素变得更细,表面的凸起减少。
南荻纤维素原料及发酵残渣的 FT-IR 和 XRD 图如 图 7 所示。
图 7 (a) 可知:酶解发酵后,3700~3000 ${\mathrm{\;{cm}}}^{-1}$ 处的吸收峰 $(- \mathrm{{OH}}$ 基团的拉伸振动) 强度增大,说明在酶解糖化过程中, 纤维素的部分氢键或共价键断裂; ${897}\left({\beta \text{-糖苷键}}\right),{1320}{\mathrm{\;{cm}}}^{-1}$${1380}{\mathrm{\;{cm}}}^{-1}$ (C-H 对称弯曲振动) [ 11 ] 处的吸收峰峰形、峰强无明显变化,说明发酵前、后南荻纤维素骨架结构无明显变化。
图 7(b)可知,酶解发酵前、后南荻纤维素的 XRD 曲线在峰形、峰数、峰位均无明显差异, 说明纤维素的结晶型无变化。但发酵前、后的南荻纤维素结晶度分别为 63.50% 和 68.18%,发酵后结晶度提升 7.37%,其原因可能是南荻纤维素无定形区被纤维素酶水解,结晶区增加。
南荻生物量大,生长区域集中,相对于传统大田作物秸秆更便于收集和储运, 且其纤维素含量比作物秸秆高 10%~15%,但是,南荻的木质纤维素结构更致密, 直接酶解发酵效率极低。基于生物质全组分高效利用策略,将南荻木质纤维素逐级分解为南荻纤维素、木质素、木糖等组分分别加以利用,可大大提升附加值。
本文的试验结果表明, 在以南荻纤维素为原料同步糖化发酵产乙醇过程中, 在底物质量分数低于 20%的情况下,乙醇含量随着底物质量分数的增加而显著增加, 但是乙醇转化率在底物质量分数为 15%时达到最大值;当底物质量分数超过 20%时,由于样品自身的吸胀性,会抑制反应体系的酶解糖化过程,体系的乙醇含量有所降低。有研究表明,在适当的时间内进行分批补料可以提高底物质量分数, 并且避免高固载带来的体系粘度高、传质效率差的缺陷, 从而促进酶解效率及乙醇转化率 [ 7 ]
正交试验结果表明,底物初始质量分数对体系的乙醇含量有极显著影响, 原因可能是纤维素酶解转化率和葡萄糖含量受底物质量分数的影响, 底物质量分数的降低能提高纤维素转化效率, 但葡萄糖含量低,不利于后续发酵产乙醇,并且会增大蒸馏成本 [ 12 ] ;但是,底物质量分数过高,纤维素吸水润胀会增大反应体系的粘稠度, 不利于体系的传质和传热效率, 也不利于纤维素酶解及酵母发酵。因此,合适的底物初始质量分数能保证反应体系从一开始就处于较优的状态, 从而保证葡萄糖产率。
补料次数对乙醇含量具有极显著影响, 这是由于补料次数越少, 反应初期南荻纤维素样品的吸胀作用导致的体系粘稠度越大, 阻碍了酶与底物的接触, 影响可发酵糖的生成, 另外体系的粘稠度高也不利于酵母的生长和发酵 [ 13 ] ;当补料次数为 3 次时,最后一次补料发生在反应 ${48}\mathrm{\;h}$ 后,此时纤维素酶的酶活有所下降, 且酵母活性会随着体系中乙醇含量、葡萄糖含量的不断增加而受到抑制。
底物最终质量分数对乙醇含量没有显著影响。在本文中,底物最终质量分数被设置为 ${30}\%$ , 32.5%和 35%,均属于较高底物质量分数范围,此时, 分批补料同步糖化发酵条件对乙醇产率无显著影响。本试验优化确立了南荻纤维素分批补料同步糖化发酵产乙醇的最佳条件,最佳条件下的乙醇含量较未分批补料试验中获得的最大乙醇含量提高了 23.36%。
物料平衡分析结果表明,在分批补料过程中, 南荻纤维素的酶解转化率大于 80%,但是乙醇转化率仅为 50.12%, 导致体系中葡萄糖剩余 102.98 g。因此,后续研究的重点在于进一步提升纤维素酶解转化效率以及乙醇的转化效率,特别是后者。
酶解发酵过程较大地改变了南荻纤维素的表观形貌, 使结构疏松、表面伴有球状、片状凸起的条状纤维素变得更细, 表面的凸起减少, 原因是凸起状纤维素的比表面积更大, 与纤维素酶最先接触,可及性更好,因此最先被消耗。南荻纤维素在纤维素酶的作用下发生逐层酶解糖化,但该过程对底物的骨架结构以及结晶型无显著性改变。
以南荻纤维素为原料, 采用同步糖化发酵的方式生产乙醇,当底物质量分数为 20% 时, 可获得的最大乙醇含量为 ${56.87}\mathrm{\;g}/\mathrm{L}$ ,当底物质量分数超过 20%时,乙醇含量会显著下降。采用分批补料的方式能提升反应体系中的乙醇含量, 采用单因素试验及正交试验优化得到分批补料同步糖化发酵的最优工艺,即底物初始质量分数为 17.5%, 在 ${12},{24}\mathrm{\;h}$ 后进行 2 次分批补料至底物最终质量分数为 32.5%,在此条件下,试验所得最大的乙醇含量为 ${70.15}\mathrm{\;g}/\mathrm{L}$ ,比未分批补料试验中获得的最大乙醇含量提高了 23.36%。物料平衡分析表明, 在此反应体系中, 限制乙醇含量进一步提升的因素主要是乙醇转化效率和纤维素的酶解糖化效率,这是今后研究的重点。酶解发酵过程会改变南荻纤维素的表观形貌, 但是对其骨架结构以及结晶型无显著性影响。
